DIFFUSE TRAUMATIC AXONAL INJURY WITHIN THE VISUAL SYSTEM: IMPLICATIONS FOR VISUAL PATHWAY REORGANIZATION By Jiaqiong Wang, Ph.D. Candidate A dissertation submitted in partial fulfillment of the requirements for the degree of Doctor of Philosophy at Virginia Commonwealth University Virginia Commonwealth University, 2012 Major Director: John T. Povlishock, Ph.D. Professor and Chairman Department of Anatomy and Neurobiology Traumatic brain injury is a major health problem with much of its morbidity associated with traumatic axonal injury (TAI). To date, significant insight has been gained into the initiating pathogenesis of TAI. However, the specific anterograde and retrograde sequelae of TAI are poorly understood because the diffuse nature of TAI complicates data analysis. To overcome this limitation, we subjected transgenic mice expressing yellow fluorescent protein (YFP) within the visual system to central fluid percussion injury, and consistently generated diffuse TAI within the optic nerve that could easily be followed in the organized YFP positive fibers. Diffuse traumatic axonal injury in the mouse induces atrophy, c-Jun activation, and axonal outgrowth in the axotomized neuronal population.
Traumatic axonal injury (TAI) is a consistent component of traumatic brain injury (TBI) and is associated with much of its morbidity. Little is known regarding the long-term retrograde neuronal consequences of TAI and/or the potential that TAI could lead to anterograde axonal reorganization and repair. To investigate the repertoire of anterograde and retrograde responses triggered by TIA, Thy1-...
متن کاملDiffuse Axonal Injury: Pathological and Clinical Aspects
Diffuse traumatic brain injury refers to widespread pattern of injury throughout the brain caused by traumatic disruption of nerve cells, with particular injury to the axons [1]. There are four principal types of traumatic brain injury: diffuse vascular injury, diffuse axonal injury, hypoxic-ischemic encephalopathy and diffuse brain swelling [2]. Clinically, DAI is characterized by generalized ...
متن کاملDiffuse axonal injury in brain trauma: insights from alterations in neurofilaments
Traumatic brain injury (TBI) from penetrating or closed forces to the cranium can result in a range of forms of neural damage, which culminate in mortality or impart mild to significant neurological disability. In this regard, diffuse axonal injury (DAI) is a major neuronal pathophenotype of TBI and is associated with a complex set of cytoskeletal changes. The neurofilament triplet proteins are...

Traumatic brain injury (TBI) is a prevalent condition and there is limited visual perception research with this population. Here, we investigated perceptual organization changes in a rather homogeneous sample of closed head TBI outpatients with diffuse axonal injury only and no other known comorbidities. Patients had normal or corrected visual acuity. Perceptual organization was measured with t...
متن کاملA Possible Role for Integrin Signaling in Diffuse Axonal Injury
Over the past decade, investigators have attempted to establish the pathophysiological mechanisms by which non-penetrating injuries damage the brain. Several studies have implicated either membrane poration or ion channel dysfunction pursuant to neuronal cell death as the primary mechanism of injury. We hypothesized that traumatic stimulation of integrins may be an important etiological contrib...
